A construct encoding NH2-terminally 6xHis-tagged mouse glucose-6-phosphate isomerase (Gpi) was expressed in the yeast cells. The resulting protein is the recombinant full-length of mature Gpi protein containing amino acid sequence 2-558 of mouse Gpi. The purity of this Gpi protein is greater than 90% determined by SDS-PAGE. A molecular mass band range from 60 kDa to 66 kDa was presented on the gel under reducing conditions. In-stock Gpi proteins are offered now. This recombinant Gpi protein may be used to produce anti-Gpi antibodies or in the studies of glycolysis, gluconeogenesis, or Gpi-related disorders.
Gpi is a glycolytic enzyme that catalyzes the interconversion of glucose-6-phosphate and fructose-6-phosphate, the second reaction step of glycolysis. It is also involved in gluconeogenesis and the pentose phosphate pathway. Besides, Gpi can be secreted to the extracellular matrix functioning like a cytokine or growth factor. Studies demonstrated that Gpi acts as a neurotrophic factor that promotes the survival of skeletal motor neurons and sensory neurons and as a lymphokine that stimulates the secretion of immunoglobulin. Gpi deficiency causes nonspherocytic hemolytic anemia and is even related to fetal edema, immediate neonatal death, and neurological dysfunction.
