IL4R
IL-4R由802個(gè)氨基酸殘基組成，分子量為140kDa，屬于紅細(xì)胞生成素受體超家族成員。其結(jié)構(gòu)具有特定的特征，包括兩個(gè)纖連蛋白結(jié)構(gòu)域（fnIII），每個(gè)結(jié)構(gòu)域由六個(gè)β片組成。在IL-4與IL-4Rα亞基結(jié)合時(shí)，這些結(jié)構(gòu)域有助于形成異二聚體。IL-4R在多種細(xì)胞過程中扮演重要角色。它參與免疫細(xì)胞的存活、發(fā)育，調(diào)控B細(xì)胞的發(fā)育和成熟，以及Ⅱ型輔助性T細(xì)胞的增殖分化。此外，IL-4R的表達(dá)與多種腫瘤細(xì)胞的存活、增殖、遷移和侵襲密切相關(guān)。在腫瘤微環(huán)境中，IL-4R的表達(dá)上調(diào)，并通過多種機(jī)制促進(jìn)腫瘤轉(zhuǎn)移。 IL-4R的生物學(xué)功能主要通過非受體型酪氨酸激酶JAK激活。當(dāng)IL-4與IL-4R結(jié)合時(shí)，它會(huì)激活JAK，進(jìn)而觸發(fā)一系列的信號(hào)級(jí)聯(lián)反應(yīng)。這些反應(yīng)包括STAT6的磷酸化和核轉(zhuǎn)位，以及其他下游信號(hào)分子的激活。這些信號(hào)通路最終影響細(xì)胞的增殖、分化、存活和遷移等過程。
