多克隆抗體

抗體通常分為兩大類(lèi),分別是單克隆抗體和多克隆抗體。顧名思義,由一種抗原決定簇刺激機(jī)體,由一個(gè)B淋巴細(xì)胞接受該抗原刺激所產(chǎn)生的抗體稱(chēng)之為單克隆抗體。由多種抗原決定簇刺激機(jī)體,相應(yīng)地就產(chǎn)生各種各樣的單克隆抗體,這些單克隆抗體混雜在一起就是多克隆抗體,機(jī)體內(nèi)所產(chǎn)生的抗體就是多克隆抗體。簡(jiǎn)言之,多克隆抗體就是用一種包含多種抗原決定簇的抗原免疫動(dòng)物,刺激機(jī)體多個(gè)B細(xì)胞克隆產(chǎn)生針對(duì)多種抗原表位的不同抗體后,所獲得的免疫血清實(shí)際上是含有多種抗體的混合物。

關(guān)于多抗

正如前面所提到的,多克隆抗體猶如許多單克隆抗體的集合,它們有的特異性高,有的特異性低,但總體特異性符合正態(tài)分布曲線的形式,而其特異性的總體表現(xiàn)也應(yīng)處于統(tǒng)計(jì)學(xué)正態(tài)曲線中位線的水平。與單克隆抗體相比,多克隆抗體的優(yōu)缺點(diǎn)如下:

優(yōu)勢(shì):

  • 由于多克隆抗體具有多個(gè)表位結(jié)合位點(diǎn),可與靶蛋白的多個(gè)表位結(jié)合,所以,多克隆抗體有助于放大低表達(dá)水平的靶蛋白信號(hào),但是,從結(jié)果的準(zhǔn)確性來(lái)說(shuō),這個(gè)優(yōu)勢(shì)也是劣勢(shì),它會(huì)給定量實(shí)驗(yàn)造成不利影響,使結(jié)果變得不準(zhǔn)確。
  • 由于能識(shí)別多個(gè)表位,多克隆抗體更適用于IP/ChIP實(shí)驗(yàn),可得到更好的結(jié)果。
  • 制備生產(chǎn)時(shí)間短,所需技術(shù)和技能要求不高,成本比單克隆抗體低,穩(wěn)定性強(qiáng)。
  • 雙抗夾心法ELISA中捕獲抗體首選多克隆抗體,可以快速捕獲靶蛋白。

劣勢(shì):

  • 易于產(chǎn)生批次間差異。
  • 可產(chǎn)生大量非特異性抗體,使應(yīng)用中背景信號(hào)強(qiáng)。
  • 多表位使得檢測(cè)免疫原序列以確定是否有任何交叉反應(yīng)變得尤為重要。
