TM4SF1

TM4SF1（Transmembrane 4 L six family member 1）是Tetraspanin超家族中亞家族TM4SFs的一員，首次于1986年被鑒定。它具有Tetraspanin家族典型的四次跨膜結(jié)構(gòu)，胞外含兩個(gè)loop區(qū)EC1和EC2，其中EC2區(qū)可通過(guò)半胱氨酸殘基與整合素α5β1結(jié)合，影響細(xì)胞的黏附和運(yùn)動(dòng)。C端胞質(zhì)區(qū)包含PDZ結(jié)合域，可與Syntenin-1和Syntenin-2蛋白相互作用介導(dǎo)腫瘤細(xì)胞的增殖和轉(zhuǎn)移。 TM4SF1在多種腫瘤細(xì)胞表面高表達(dá)，包括肺癌、乳腺癌、肝癌、結(jié)直腸癌及卵巢癌等，且能與脂質(zhì)激酶、酪氨酸蛋白激酶等形成復(fù)合體，如可結(jié)合DDR1，激活PKC和JAK2-STAT3信號(hào)通路，促進(jìn)細(xì)胞偽足延伸。初步的研究結(jié)果表明，TM4SF1作為具有酪氨酸蛋白激酶活性的單次跨膜受體1（DDR1）的輔助受體，調(diào)控下游的PKC/STAT3信號(hào)通路，從而影響乳腺癌向肺組織的轉(zhuǎn)移。此外，TM4SF1也是具有新型內(nèi)化途徑的ADC靶點(diǎn)，這意味著它在抗體偶聯(lián)藥物（ADC）的設(shè)計(jì)和開(kāi)發(fā)中可能具有潛在的應(yīng)用價(jià)值。 于TM4SF1的藥物研發(fā)處于初級(jí)階段，尚未有成熟的藥物應(yīng)用于臨床治療。因此，對(duì)于TM4SF1的研究仍具有深遠(yuǎn)的意義和廣闊的前景。
