STAT3

STAT3，即信號轉(zhuǎn)導和轉(zhuǎn)錄激活因子3，別名包括急性調(diào)節(jié)蛋白、信號轉(zhuǎn)導激活因子、亞油酸誘導蛋白等。STAT3是一種轉(zhuǎn)錄因子，廣泛存在于多種細胞類型中，由人體細胞自身的基因編碼。其作用機制主要是在細胞接收到外部信號，如細胞因子、生長因子或其他刺激后，被激活的酪氨酸激酶磷酸化，形成二聚體，隨后轉(zhuǎn)移到細胞核內(nèi)調(diào)控特定基因的表達。STAT3的激活涉及JAK/STAT、IL-6/STAT3等信號通路，其中Janus激酶（JAK）首先被激活并磷酸化STAT3，導致其同源二聚化和核內(nèi)轉(zhuǎn)位。STAT3在生物學上的意義非常廣泛，它參與調(diào)節(jié)細胞的多種生理過程，包括細胞增殖、分化、遷移、凋亡以及免疫應(yīng)答等。STAT3的激活對于維持細胞穩(wěn)態(tài)和適應(yīng)外界變化至關(guān)重要。此外，STAT3在多種疾病的發(fā)生發(fā)展中也扮演著關(guān)鍵角色，尤其是在腫瘤發(fā)展中，STAT3的持續(xù)激活與腫瘤細胞的增殖、存活、血管生成和免疫逃逸等密切相關(guān)。因此，STAT3成為了癌癥治療中的一個重要靶點。在自身免疫性疾病、炎癥反應(yīng)和組織修復中，STAT3的調(diào)節(jié)作用同樣不容忽視，靶向STAT3的干預(yù)策略可能為這些疾病的治療提供新的途徑。
