ROS1

ROS1,即c-ros肉瘤致癌因子-受體酪氨酸激酶,別名CD74-ROS1、EZR-ROS1、SLC34A2-ROS1等,這些別名通常表示ROS1基因與其他基因發(fā)生融合的形式。ROS1基因定位于人類6號(hào)染色體長臂,屬于酪氨酸激酶胰島素受體基因,由胞內(nèi)激酶區(qū)、跨膜區(qū)及胞外區(qū)三部分組成,編碼具有酪氨酸激酶活性的跨膜蛋白 。ROS1的作用機(jī)制涉及其作為細(xì)胞表面受體的功能,當(dāng)接收到外部特定信息后活化,并傳遞信號(hào)到細(xì)胞內(nèi)部,指導(dǎo)細(xì)胞有序地生長和增殖。若ROS1基因發(fā)生致癌變異,會(huì)激活其下游通路的信號(hào),造成細(xì)胞過度地生長及增殖,驅(qū)動(dòng)腫瘤的發(fā)生。ROS1主要參與的信號(hào)通路包括RAS-RAF-MEK-ERK(MAPK)、PI3K-AKT-mTOR等 。在生物學(xué)意義上,ROS1基因融合在多種腫瘤中被認(rèn)為是致癌因素,特別是在非小細(xì)胞肺癌中,ROS1融合基因型肺癌的發(fā)病率較低,但具有獨(dú)特的臨床病理特征。ROS1融合基因的發(fā)現(xiàn)為靶向治療提供了新的治療靶點(diǎn),已有多種針對ROS1的小分子抑制劑在臨床試驗(yàn)中顯示出療效,如克唑替尼等 。

Immunofluorescence staining of Hela cells with CSB-PA020072LA01HU at 1:133, counter-stained with DAPI. The cells were fixed in 4% formaldehyde, permeabilized using 0.2% Triton X-100 and blocked in 10% normal Goat Serum. The cells were then incubated with the antibody overnight at 4°C. The secondary antibody was Alexa Fluor 488-congugated AffiniPure Goat Anti-Rabbit IgG(H+L).
