NFE2L2

NFE2L2,即核因子(紅細胞衍生2)相關(guān)因子2,別名NRF2、NFE2L2。它是一種轉(zhuǎn)錄因子,由人體細胞自身的基因編碼,在細胞防御氧化應(yīng)激和清除有害物質(zhì)中發(fā)揮關(guān)鍵作用。NFE2L2的作用機制涉及多個層面,包括翻譯后修飾、蛋白相互作用和表觀遺傳調(diào)控等。在正常生理條件下,NFE2L2與KEAP1結(jié)合并通過泛素-蛋白酶體途徑促進其降解,維持NFE2L2表達在較低水平。當細胞遭遇氧化應(yīng)激或親電試劑時,NFE2L2會從細胞質(zhì)轉(zhuǎn)移到細胞核,與抗氧化反應(yīng)元件(ARE)結(jié)合,激活抗氧化相關(guān)基因的轉(zhuǎn)錄。NFE2L2在生物學(xué)上的意義非常廣泛,它參與調(diào)控黃曲霉毒素分解代謝過程、大分子代謝過程的正向調(diào)控以及細胞遷移的調(diào)節(jié)。NFE2L2的功能異常與多種疾病相關(guān),包括糖尿病視網(wǎng)膜病變、肝硬化、大腦中動脈梗塞、非酒精性脂肪肝病等。此外,NFE2L2的人類直系同源基因與阿爾茨海默病、白內(nèi)障、肝細胞癌、黃斑變性、肺結(jié)核等疾病有關(guān)。在癌癥中,NFE2L2-KEAP1信號通路經(jīng)常被劫持,尤其是在非小細胞肺癌中,NFE2L2的激活可導(dǎo)致癌細胞逃脫各種抗腫瘤治療,促進疾病進展和轉(zhuǎn)移。因此,NFE2L2是細胞氧化應(yīng)激反應(yīng)和疾病治療中的一個重要靶點。

Immunofluorescence staining of Hela Cells with CSB-RA225569A0HU at 1:50, counter-stained with DAPI. The cells were fixed in 4% formaldehyde, permeated by 0.2% TritonX-100, and blocked in 10% normal Goat Serum. The cells were then incubated with the antibody overnight at 4℃. Nuclear DNA was labeled in blue with DAPI. The secondary antibody was FITC-conjugated AffiniPure Goat Anti-Rabbit IgG (H+L).
