NECTIN4

脊髓灰質(zhì)炎病毒受體4（Nectin-4）屬于免疫球蛋白超家族（IgSF），是一類新發(fā)現(xiàn)的細(xì)胞黏附分子。NECTIN4作為一種I型膜蛋白，在正常的胚胎和胎兒組織中含量很高，成年后下降，在健康組織中的分布非常有限。然而，Nectin-4蛋白在尿路上皮癌、乳腺癌、膀胱癌、卵巢癌、胃癌、肝細(xì)胞癌和胰腺癌等組織中都具有特異性的高表達(dá)，其主要通過激活PI3K/Akt途徑促進(jìn)腫瘤細(xì)胞增殖、分化、遷移、侵襲等。目前，已上市的Nectin-4 ADC藥物Padcev（Enfortumab vedotin）已展示出與免疫檢查點(diǎn)抑制劑聯(lián)用的優(yōu)異臨床潛力，用于局部晚期或轉(zhuǎn)移性尿路上皮癌成人患者。未來，靶向Nectin-4的抗體-藥物偶聯(lián)物有望進(jìn)一步將適應(yīng)癥拓寬至乳腺癌和胃癌等其它惡性腫瘤。
