GCGR

胰高血糖素受體(Gprotein-coupledreceptor, GCGR)屬于B類G蛋白偶聯(lián)受體家族(GPCRs),在維持人體血糖穩(wěn)態(tài)中發(fā)揮關(guān)鍵作用。其它相關(guān)的G-蛋白偶聯(lián)受體還包括胰高血糖素-樣肽-1受體(GLP-I)、胰高血糖素-樣肽-2受體(GLP-2)和胃抑制性多肽受體等。GCGR主要在肝臟和腎臟中表達,在心臟,脂肪組織,脾臟,胸腺,腎上腺,胰腺,大腦皮層和胃腸道中有少量發(fā)現(xiàn)。GCGR的天然配體為胰高血糖素(Glucagon),在體內(nèi),GCGR與其配體結(jié)合,主要通過激活Gs蛋白行使其功能。激活的GCGR通過與異源三聚體Gs結(jié)合,誘導(dǎo)腺苷酸環(huán)化酶活化,從而產(chǎn)生cAMP,激活蛋白激酶A,使肝糖原分解和糖異生增加,導(dǎo)致體內(nèi)血糖升高。研究表明,針對GCGR靶點的特異性抗體可促進1型糖尿病動物的β細胞再生,改善1型糖尿病患者治療。陸續(xù)有研究報道,GCGR還與腎臟、脂肪肝、肥胖等代謝疾病相關(guān)。目前,GCGR已成為抵抗糖尿病的重要靶點。
