FSHR

卵泡刺激素受體(FSHR)屬于G蛋白偶聯(lián)受體超家族中的糖蛋白亞家族成員,由胞外區(qū)、胞內(nèi)區(qū)及跨膜區(qū)3部分組成。FSHR通過與其配體——卵泡刺激素(FSH)結(jié)合參與信號轉(zhuǎn)導(dǎo)。

FSH是垂體分泌的一種糖蛋白激素,主要作用是促進和維持正常的性腺發(fā)育和生殖功能。FSH可以通過與FSHR結(jié)合來刺激女性卵巢中卵泡的發(fā)育,促進男性睪丸的精子發(fā)生。FSH主要通過FSHR介導(dǎo)的腺苷酸環(huán)化酶-環(huán)磷酸腺苷(cAMP)途徑發(fā)揮作用。已證實FSH通過特異性結(jié)合FSHR胞外區(qū)的富含亮氨酸重復(fù)序列來激活穩(wěn)定態(tài)受體?;罨腇SHR刺激位于細胞膜內(nèi)膜的偶聯(lián)鳥苷結(jié)合蛋白(Gs蛋白)傳遞信息。被活化的Gs蛋白激活腺苷酸化話梅促成胞質(zhì)內(nèi)第二信使cAMP生產(chǎn),cAMP再激活蛋白激酶A,促進蛋白磷酸化,實現(xiàn)生理效應(yīng)。

FSHR的表達被認為是性腺特異性的。長期以來,人們認為FSHR的表達僅限于卵巢的顆粒細胞和睪丸的支持細胞,但后來發(fā)現(xiàn)FSHR在胎盤、子宮、前列腺、骨組織和卵巢上皮細胞以及卵巢癌。此外,最近有研究發(fā)現(xiàn)FSHR在許多腫瘤血管表面選擇性表達,并且發(fā)現(xiàn)FSHR的表達與原發(fā)性腎細胞癌的腫瘤轉(zhuǎn)移密切相關(guān)。FSHR的表達水平在女性卵泡發(fā)育的不同階段也不同。
