Claudin 4( CLDN4 )是一種緊密連接蛋白，是Claudin 家族的一員，Claudin 蛋白均由4個(gè)跨膜結(jié)構(gòu)域構(gòu)成，N端和C端均位于細(xì)胞內(nèi)部，細(xì)胞膜外有2個(gè)長(zhǎng)度不等的環(huán)，可以和臨近其他同類型的環(huán)相接觸構(gòu)成細(xì)胞間的緊密連接． 而且CLDN4的結(jié)構(gòu)高度保守，從線蟲到人類都十分相似．當(dāng)Claudin蛋白發(fā)生變化時(shí)將影響細(xì)胞間的通透性進(jìn)一步引起多種疾病的發(fā)生．研究報(bào)道CLDN4 在卵巢癌、前列腺癌、乳腺癌、胰腺癌、腸道癌等多種腫瘤中表達(dá)上調(diào)，并且作為一種癌基因介導(dǎo)癌癥的發(fā)生。

Then 10% normal goat serum was Incubated to block non-specific protein-protein interactions followed by the antibody (1μg/1*106cells) for 45 min at 4°C. The secondary antibody used was FITC-conjugated Goat Anti-Mouse IgG(H+L) at 1/200 dilution for 35 min at 4°C. Isotype control antibody (green line) was mouse IgG1 (1μg/1*106cells) used under the same conditions. Acquisition of >10,000 events was performed.
