共刺激/共抑制雙重作用:B7-H3是“敵”是“友”?

B7-H3(B7 homolog 3 protein)又名CD276,是由316個氨基酸組成的I型跨膜糖蛋白,分子量為45-66 kDa,其與B7-H1(PD-L1)有相似的分子結構,是B7-CD28家族重要的免疫檢查點分子。B7-H3主要以膜蛋白和可溶形式存在,可溶的B7-H3(soluble B7-H3, sB7-H3)由膜蛋白經(jīng)金屬蛋白酶剪切而來。此外,在外泌體和其他胞外小泡中也發(fā)現(xiàn)了B7-H3蛋白。B7-H3在免疫系統(tǒng)中可能發(fā)揮雙重作用。一方面,B7-H3作為一種共刺激分子,對CD4+和CD8+細胞具有共刺激作用,誘導細胞免疫,并在T細胞受體信號傳導下選擇性增強干擾素-γ(IFNG)的產(chǎn)生。另一方面,在人類和小鼠中的實驗證明,B7-H3也具有共抑制作用[10],它可以抑制Treg細胞,從而使腫瘤逃逸免疫反應。這可能與NFAT、NF-κB和AP-1因子相關。已有研究證實B7-H3在膀腺癌、前列腺癌、黑色素瘤等腫瘤細胞系中的高表達被證明可促進腫瘤細胞的侵襲和轉(zhuǎn)移。在黑素瘤細胞中,沉默B7-H3可降低細胞的基質(zhì)凝膠侵襲能力。這可能是因為B7-H3沉默可降低與轉(zhuǎn)移相關的蛋白的表達,例如基質(zhì)金屬蛋白酶(MMP)-2,信號轉(zhuǎn)導和轉(zhuǎn)錄激活因子3(STAT3)以及分泌的白介素8(IL-8)的水平。
