ACE2

血管緊張素轉(zhuǎn)化酶2(ACE2),也稱為ACEH,一種金屬蛋白酶,全長805個氨基酸,是具有單一胞外催化結(jié)構(gòu)域的I型跨膜糖蛋白。ACE2組織分布具有器官特異性,主要表達于腎臟、心血管及胃腸道系統(tǒng),在正常肺組織中,Ⅰ型及Ⅱ型肺泡上皮細胞中均存在ACE2。ACE2不僅在心血管系統(tǒng)中發(fā)揮了重要的功能,而且作為SARS-CoV-2入胞的受體在肺部疾病中發(fā)揮了一定作用。研究已經(jīng)進一步證實:對于SARS-CoV-2而言,刺突蛋白(S蛋白)是介導(dǎo)病毒入侵宿主細胞的關(guān)鍵蛋白。SARS-CoV-2是通過S蛋白與ACE2結(jié)合入侵肺部細胞。ACE2與新冠病毒的RBD(受體結(jié)合結(jié)構(gòu)域)之間的親和力,比其與SARS病毒的RBD的親和力高10到20倍。隨著ACE2為SARS-CoV-2入侵人體靶點的揭露,阻斷病毒和 ACE2的結(jié)合成為了治療方向之一。
