    • IHC image of CSB-MA192596 diluted at 1:100 and staining in paraffin-embedded human tonsil tissue performed on a Leica BondTM system. After dewaxing and hydration, antigen retrieval was mediated by high pressure in a citrate buffer (pH 6.0). Section was blocked with 10% normal goat serum 30min at RT. Then primary antibody (1% BSA) was incubated at 4°C overnight. The primary is detected by a Goat anti-mouse IgG polymer labeled by HRP and visualized using 0.05% DAB.

    After immunizing mice with a synthesized peptide derived from human KRT16, B cells are isolated from the mouse spleen and fused with myeloma cells to create hybridomas. The hybridomas are screened to select the clones that produce KRT16 antibodies. The selected hybridomas are then injected into the mouse abdominal cavity for culture, and the KRT16 monoclonal antibody is affinity-purified from mouse ascites using a specific immunogen. This results in a mouse monoclonal antibody that specifically reacts with human KRT16 protein in ELISA and IHC applications.

    KRT16 is a type I intermediate filament protein that is mainly expressed in hair follicles, nails, and the oral epithelium. It primarily provides structural support to cells and tissues. KRT16 forms heterodimers with other type I and type II keratin proteins and assembles into intermediate filaments that are important for maintaining the integrity of epithelial cells. Mutations in the KRT16 gene have been associated with various skin and nail disorders, including pachyonychia congenita.
