MICA

MICA(MHC類I鏈相關(guān)蛋白A)是位于人類MHC區(qū)域的非經(jīng)典MHC I類分子,具有高度多態(tài)性。它編碼一個(gè)含有3個(gè)α螺旋的跨膜蛋白,其結(jié)構(gòu)與經(jīng)典MHC I類分子相似但存在關(guān)鍵差異。MICA的主要功能是作為應(yīng)激誘導(dǎo)分子在多種上皮腫瘤細(xì)胞表面表達(dá),并激活通過(guò)與NKG2D受體結(jié)合的免疫細(xì)胞,如NK細(xì)胞、NKT細(xì)胞、γδT細(xì)胞和CD8+T細(xì)胞,介導(dǎo)這些細(xì)胞的活化并殺傷靶細(xì)胞。MICA參與的信號(hào)通路主要是NKG2D信號(hào)通路,它通過(guò)與NKG2D受體的相互作用觸發(fā)免疫細(xì)胞的細(xì)胞毒性程序和共刺激信號(hào)通路,對(duì)腫瘤細(xì)胞逃脫機(jī)體免疫監(jiān)視過(guò)程中發(fā)揮主要作用。腫瘤細(xì)胞可能通過(guò)蛋白水解使細(xì)胞表面MICA脫落來(lái)逃避免疫識(shí)別。MICA的功能性結(jié)構(gòu)域?yàn)榘獾?alpha;1和α2結(jié)構(gòu)域,這些結(jié)構(gòu)域?qū)γ庖呒?xì)胞的識(shí)別和激活至關(guān)重要。MICA的表達(dá)和功能對(duì)于維持機(jī)體對(duì)腫瘤的免疫監(jiān)視非常重要,其異常表達(dá)或功能失調(diào)與多種腫瘤的發(fā)生發(fā)展有關(guān)。MICA的研究有助于開(kāi)發(fā)新的癌癥治療策略,尤其是在利用免疫系統(tǒng)對(duì)抗腫瘤方面顯示出重要性。

IHC image of CSB-RA574653A0HU diluted at 1:80 and staining in paraffin-embedded human small intestine tissue performed on a Leica BondTM system. After dewaxing and hydration, antigen retrieval was mediated by high pressure in a citrate buffer (pH 6.0). Section was blocked with 10% normal goat serum 30min at RT. Then primary antibody (1% BSA) was incubated at 4°C overnight. The primary is detected by a Goat anti-rabbit polymer IgG labeled by HRP and visualized using 0.49% DAB.
