ICAM1

ICAM1,即細(xì)胞間黏附分子1,別名包括CD54。它是一種由ICAM1基因編碼的細(xì)胞表面糖蛋白,通常在內(nèi)皮細(xì)胞和免疫系統(tǒng)的細(xì)胞上表達(dá)。ICAM1的作用機制涉及與白細(xì)胞相關(guān)抗原1(LFA-1)和巨噬細(xì)胞1(Mac-1)等整合素家族成員的相互作用,這些整合素是ICAM-1的主要受體。ICAM1在多種生物學(xué)過程中發(fā)揮作用,包括細(xì)胞黏附、信號轉(zhuǎn)導(dǎo)、免疫應(yīng)答、炎癥反應(yīng)、細(xì)胞增殖、分化、遷移和凋亡等。ICAM1參與了多種生理和病理過程的調(diào)節(jié)。ICAM1的表達(dá)在炎癥反應(yīng)中增加,促進(jìn)白細(xì)胞與內(nèi)皮細(xì)胞的黏附,有助于白細(xì)胞穿越血管壁進(jìn)入炎癥部位。此外,ICAM1在控制腫瘤惡化和轉(zhuǎn)移以及調(diào)節(jié)機體免疫反應(yīng)中也起重要作用。ICAM1通過與其受體的特異性結(jié)合,增強白細(xì)胞、炎癥細(xì)胞和腫瘤細(xì)胞等與內(nèi)皮細(xì)胞間的黏附作用,促進(jìn)內(nèi)皮細(xì)胞活化,使它們更容易穿透內(nèi)皮。ICAM1還參與細(xì)胞的信號轉(zhuǎn)導(dǎo)與活化、細(xì)胞組織生長及分化、血管生成及腫瘤轉(zhuǎn)移等生理病理過程 。

IHC image of CSB-RA162789A0HU diluted at 1:100 and staining in paraffin-embedded human tonsil tissue performed on a Leica BondTM system. After dewaxing and hydration, antigen retrieval was mediated by high pressure in a citrate buffer (pH 6.0). Section was blocked with 10% normal goat serum 30min at RT. Then primary antibody (1% BSA) was incubated at 4℃ overnight. The primary is detected by a Goat anti-rabbit IgG polymer labeled by HRP and visualized using 0.05% DAB.

The cells were incubated in 10% normal goat serum to block non-specific protein-protein interactions followedby the antibody (1μg/1*106 cells) for 1 h at 4℃.The secondary antibody used was FITC-conjugated goat anti-rabbit IgG (H+L) at 1/200 dilution for 30min at 4℃. Control antibody (green line) was Rabbit IgG (1μg/1*106 cells) used under the same conditions. Acquisition of >10,000 events was performed.
