GNRHR

GNRHR，即促性腺激素釋放激素受體，別名GRHR、Gonadotropin-releasing hormone receptor、GnRH receptor、GnRH-R。它是一種G蛋白偶聯(lián)受體，主要在垂體前葉中表達(dá)，參與調(diào)節(jié)生殖激素的釋放。GNRHR的作用機(jī)制主要是通過與促性腺激素釋放激素（GnRH）結(jié)合，激活受體，進(jìn)而通過G蛋白介導(dǎo)的信號(hào)轉(zhuǎn)導(dǎo)，增加細(xì)胞內(nèi)cAMP的水平，促進(jìn)黃體生成素（LH）和卵泡刺激素（FSH）的釋放。GNRHR不僅在生殖系統(tǒng)中發(fā)揮關(guān)鍵作用，還與多種疾病的發(fā)生發(fā)展有關(guān)。例如，在乳腺癌中，GNRHR的表達(dá)與腫瘤的臨床病理特征和預(yù)后相關(guān)，特別是在激素受體陰性的乳腺癌中，GNRHR的高表達(dá)可能與改善的無病生存期（DFS）相關(guān)。此外，GNRHR的信號(hào)傳導(dǎo)在垂體細(xì)胞中涉及絲裂原活化蛋白激酶（MAPK）級(jí)聯(lián)的激活，為信號(hào)從細(xì)胞表面到細(xì)胞核的傳遞提供了重要的聯(lián)系，并在促性腺激素的轉(zhuǎn)錄調(diào)節(jié)中起作用。GNRHR也在非垂體細(xì)胞中表達(dá)，并發(fā)現(xiàn)其在某些細(xì)胞中抑制細(xì)胞增殖，因此，GNRHR代表了用于癌癥治療的GnRH類似物的潛在靶標(biāo)。
