    This mouse LAG3 ELISA kit employs the quantitative sandwich enzyme immunoassay technique to measure the levels of mouse LAG3 in multiple samples, including serum, plasma, or tissue homogenates. It also uses the enzyme-substrate chromogenic reaction to visualize and analyze the analyte levels through the color intensity. The intensity of the colored product is in direct proportion to the LGA3 levels in the sample and is measured at 450 nm through a microplate reader.

    LAG3 is mainly expressed in activated T and natural killer (NK) cells and was identified as a marker for the activation of CD4+ and CD8+ T cells. Under pathological conditions, such as chronic inflammation or tumor environment, increased LAG3 expression on T cells was observed in combination with other inhibitory receptors such as PD-1, T cell immunoglobulin, and TIGIT, TIM3, CD160, 2B4, which finally led to T cell dysfunction. LAG3 is an important immune checkpoint with relevance in cancer, infectious disease, and autoimmunity. Furthermore, LAG3 was mainly found on tumor-infiltrating Tregs in many types of cancer when compared with non-malignant peripheral cells.

    Lymphocyte activation gene 3 protein: Inhibitory receptor on antigen activated T-cells. Delivers inhibitory signals upon binding to ligands, such as FGL1. FGL1 constitutes a major ligand of LAG3 and is responsible for LAG3 T-cell inhibitory function. Following TCR engagement, LAG3 associates with CD3-TCR in the immunological synapse and directly inhibits T-cell activation. May inhibit antigen-specific T-cell activation in synergy with PDCD1/PD-1, possibly by acting as a coreceptor for PDCD1/PD-1. Negatively regulates the proliferation, activation, effector function and homeostasis of both CD8(+) and CD4(+) T-cells. Also mediates immune tolerance: constitutively expressed on a subset of regulatory T-cells (Tregs) and contributes to their suppressive function. Also acts as a negative regulator of plasmacytoid dendritic cell (pDCs) activation.
